A STUDY OF THE EFFECT OF PROSTAGLANDIN E, ON THE SKIN BLOOD FLOW OF RABBITS

Abstract
|
|
|
The prostaglandin are a family of drugs with many actions. One compound, prostaglandin E, (PG ; ) has been shown to increase peripheral blood flow in animal and human studies. PGE, ha:, been used in the treatment of both Raynaud¢¥s disease and peripheral arterial occlusion -hat are not amenable to surgical reconstruction. Clinical results has been inconclusive.
We studied the prostaglandin E, effect on the nutrient skin blood flow of normal rabbits and infused intravenously PGE, (50ng/kg/min) into the ear vein.
For the measurement of skin blood flow, we used the isotope clearance technique with technetium-99m. Bi-exponential curves were always obtained from the scapular and knee area of rabbits.
The results obtained were summarized as follow;
1. The normal skin blood flow of a rabbit was 4.7¡¾ 1.3 m1/min /100 gm in scapula and 2.6¡¾0.8ml/min/i+_iOgm in knee.
2. In flurb orofen group which was a strong inhibitor to prostaglandin synthesis, the skin blood flow si owed Rio significant change from controls.
3. In prostaglandin E, group, the skin blood flow revealed much increase compared with controls.; 26.2¡¾4.3m1/miu/100 gm in scapula and 8.8¡¾1.8m1/min/IOOgm in knee.
4. At the evaluation of the survival area in random pattern skin flap (4x 12cm), the flurbiprofen grout) revealed 8.1% increase and the prostaglandin E, group showed 50.8% increase compared with the control group.
We conclude ¢¥hat PGE, increases absolute nutrient skin blood flow and the survival rate of skin flap.
